Gameplay will be similar to the first Super Mario Galaxy, with a focus on platforming based on and around 3D planets with varying sizes and landscapes. Power-ups such as the Bee Suit will be returning, along with the Launch Star for interplanetary navigation and the Airships operated by the villain Bowser and his son, Bowser Jr.

The protagonist Mario will be able to ride the bipedal dinosaur Yoshi, who can use his tongue to swing across gaps, as well as eat different fruits that give him different powers, such as the ability to inflate like a balloon, as well as speed boosts. While the player retains the ability to collect Star Bits with the blue Star Pointer from the first game, while riding Yoshi the Star Pointer is replaced by a red dot, which allows the player to also aim and control his tongue. This can be used for swinging off objects, pulling levers or, like previous games, swallowing enemies and spitting them out.

New items and gameplay mechanics were also showcased at E3 2009. These include a drill that allows Mario to dig through dirt into new areas (and in some cases explore the interior of planets) and switches that slow time momentarily, indicated by a change to monochrome vision. Players can collect comet medals hidden in the stages to unlock harder levels with tougher challenges.

Metroid: Other M is a video game in development by Team Ninja and Nintendo SPD Production Group 1 (collectively called "Project M) for the Wii. It was announced by Nintendo of America president and CEO Reggie Fils-Aime and a trailer was briefly shown during the Electronic Entertainment Expo 2009. The game features both 2D and 3D gameplay in both first- and third-person perspectives. Fils-Aime stated that Metroid: Other M would "take you deeper into Samus's story", and also noted that the game would be a return to the style of the traditional series as opposed to the Prime series, though the game would have a "harder edge". A demo was released at the Nintendo Media Summit in San Francisco.

Line Attack Heroes is a fast-paced action game in which you follow a bunch of heroes on a quest to find missing pieces of a Dragon Statue. When you defeat an enemy, they become a part of your character's attack line and can be used to defeat more enemies. Rather than simply attacking, you will command your line of warriors to attack for you.

Transformers: War for Cybertron challenges players to become the ultimate weapon as a Transformers character in the final, epic war that will determine the survival of their entire race. Complete with several multiplayer modes, Transformers: War for Cybertron allows gamers to play through story missions with their friends in drop in/drop out online co-op, and also create their own Transformers character for competitive head-to-head multiplayer modes, choosing among four distinct character classes, personalizing its look and selecting from a huge variety of weapons, skills and abilities.

A heroic tale of redemption and discovery, Epic Mickey is an action-platforming game featuring the iconic Mickey Mouse in a fantastical adventure inspired by the cartoons of the 1930s and 1940s. Pulled into a warped Disney universe called the Cartoon Wasteland by an evil villain, Mickey finds himself in a world inhabited by the retired and forgotten cartoon characters and attractions from Disney's past. Mickey must use the very elements of himself and the world, paint and paint thinner, to explore the Cartoon Wasteland, find the source of evil that is destroying the land, and redeem his fellow cartoon characters including Oswald the Lucky Rabbit, Walt Disney's first cartoon star and Mickey's resentful half-brother. With visionary game designer Warren Spector behind the title, players choose their own path to defeat supreme evil, face the consequences of those decisions and ultimately free the Cartoon Wasteland.
